(note that not all games are in a 'working' state, so pick a 'working' one. Browse to the folder with the mame.exe, then open mame.exe and tell it what folder holds all your roms, then try opening one. This will tell you where the problem lies. I had to do this manually for each sub folder. First thing to check is if you can launch the game (s) in MAME itself, without involving Launchbox.
